The principle of enzyme-linked immunosorbent assay (ELISA) is to detect the absorbance value of the tested substance at a specific wavelength. The difference in energy before and after light passing through the object being tested is the energy absorbed by the object being tested. At a specific wavelength, the concentration of the same object being tested is quantitatively related to the absorbed energy. The testing unit is represented by OD value, OD stands for optical density, OD=1og (1/trans), where trans is the transmittance value of the detected substance. According to Lambert Beer’s law, The OD value is related to the light intensity as follows: E=OD=log Ⅰ 0/Ⅰ, where E represents the absorbed light density; 0 is the light intensity before the detection object; The intensity of light coming out of the object being tested.
The OD value is calculated by the following formula: E=OD=C x D x E, where C is the concentration of the detected substance; D is the thickness of the test object; E is the molar factor.
